Neurite outgrowth assay using CELLAZIP™

This application note demonstrates neurite outgrowth assays using three types of iPS-derived neurons — DopaNeurons, GABANeurons, and Motor Neurons — in CELLAZIP™ 384-well plates. Researchers evaluated the effects of rotenone and colchicine on neurite growth and generated dose-response curves using MAP2 immunostaining and Yokogawa CQ1 imaging.

Key findings:

  • CELLAZIP™ 384-well plates supported clear, high-quality imaging across all three neuron types
  • Both rotenone and colchicine inhibited neurite outgrowth in a concentration-dependent manner across DopaNeurons, GABANeurons, and Motor Neurons
  • Colchicine inhibited neurite outgrowth at lower concentrations than rotenone across all neuron types
  • Dose-response curves were successfully generated for both compounds across a 0–20 µM range
  • CELLAZIP™ plates can be stored at room temperature for up to 12 months before use
